Advances in RNA sequencing are revealing that much of cellular regulation occurs beyond protein-coding mRNA, within diverse non-coding RNA populations that are largely missed by traditional single-cell methods. TotalX extends standard 10x Genomics workflows to capture both coding and non-coding RNA at scale, enabling simultaneous measurement of mRNA, microRNA, tRNA, and other regulatory transcripts in thousands of individual cells. By integrating this expanded RNA landscape into familiar analysis pipelines, TotalX uncovers cell-type-specific regulatory modules, clarifies microRNA–mRNA interactions, enables detection of non-polyadenylated viral RNA during infection, and reveals dynamic non-coding RNA programs during human brain development. Together, these capabilities demonstrate how expanding RNA sequencing beyond polyA transcripts provides a more complete and biologically informative view of cellular state and regulation.
