do you add any extra lysis step/enhancers during sample prep/extraction so as to optimise yield as well as recover rare RNA populations for RNA-seq? Short sequences such as the S100 proteins and secretory immunoglobulins can be lost to extraction if particular attention is not given to them due to their lightness and short sequences and they are very important in understanding complement cascade as well as rare genetic conditions/silent but important pathogen and host mutations.
