Zymo Research announces early access to the first total RNA-Seq library prep kit with fully-integrated universal rRNA depletion.
Total RNA-Seq analysis enables researchers to analyze differential expression, novel transcripts, splicing and isoforms, SNP expression, and gene fusion and other mutation events, all from the same experiment. Current methods, however, are long (6.5-10.5 hrs), complex (>100 pipetting steps), require additional customization of specialized kits/modules for ribosomal RNA and/or other abundant transcript removal (e.g. Globin), which introduces bias into absolute transcript profiling. The Zymo-Seq RiboFree™ Total RNA Library Prep takes 3.5 hours to complete and works with any total RNA sample with little-to-no bias (Figure 1). It’s RNA-Seq, made simple.

Alternate probe-based methods such as TruSeq® Ribo-Zero™ Gold (Illumina®) deplete a significant number of non-target genes. Even greater bias is found in mRNA-Seq, as a result of poly(A) enrichment upstream of library preparation, leading to a distorted view of gene expression.
