Understanding how Mycobacterium tuberculosis (M. tuberculosis) responds to drug treatments at the molecular level is crucial for improving tuberculosis (TB) therapies. Researchers at the National Microbiology Laboratory in Canada present a refined RNA sequencing (RNA-seq) method designed to analyze gene expression in M. tuberculosis with high precision.

The researchers tested their protocol on 11 different clinical strains of M. tuberculosis, with 12 replicates per strain, both in the presence and absence of the anti-TB drug pyrazinamide. Their approach combined mechanical cell lysis, TRIzol™ phase separation, and column-based purification, ensuring high-quality RNA extraction from bacterial cultures. By depleting ribosomal RNA and preparing complementary DNA (cDNA) libraries, they optimized transcript detection at a sequencing depth of 15 million reads.

The study successfully identified differences in gene expression between treated and untreated samples, confirming the method’s suitability for RNA-seq transcriptomics in M. tuberculosis research. These findings could help researchers better understand drug resistance mechanisms and develop more effective treatments against TB.
